Is it possible to make a single UFD with the ability to choose one of several USB layouts when booting to TBWinPE? Or do I need to have a dedicated UFD for each USB layout I may need to use?

You would need to create a separate UFD for each layout. They are different formatting/configurations and can't be combined on one drive.
Some systems will only boot certain layouts. Depending on your systems, if you test them you may find one layout that boots on all of them. For TBWinPE you'd normally use one of the partition layouts. For example: Partition - MBR FAT/FAT32 Partition, Partition - MBR FAT/FAT32 Partition (Int13h Extensions), etc.
